Drug intelligence / Profile preview
[18F]AlF-NOTA-A6p-FS2 is a novel fluorine-18-labeled radiopharmaceutical probe developed by the Sun Yat-sen University Cancer Center for positron emission tomography (PET) imaging of integrin α6-expressing tumors. The probe is based on a cyclic peptide (A6p) targeting the integrin α6 receptor, which is recognized as a marker for functional stem cells in multiple tumors and correlates with poor prognosis. To enhance tumor uptake and retention, the cyclic peptide was covalently modified with a fluorosulfonyl (FS) group at a specific site (FS2) and conjugated to a NOTA chelator, allowing for one-step radiolabeling with aluminum fluoride-18 ([18F]AlF). In preclinical and preliminary clinical evaluations, [18F]AlF-NOTA-A6p-FS2 demonstrated high radiochemical purity, excellent stability, and superior tumor-to-background contrast in patients with integrin α6-positive nasopharyngeal carcinoma compared to 18F-FDG.
